Summary

This NIST study addresses the analytical gap in measuring vitamin D metabolites in foods by assigning certified reference values for vitamin D2, vitamin D3, and 25-hydroxyvitamin D3 in five food-matrix Standard Reference Materials using isotope dilution LC–MS/MS. The resulting SRMs enable laboratories to validate analytical methods and compare vitamin D measurements across food sources, supporting improved understanding of the relationship between dietary vitamin D intake and nutritional status.

Regional applicability

These NIST Standard Reference Materials are used internationally by analytical laboratories, including those in the United Kingdom, to validate measurement methods and ensure comparability of vitamin D quantification in food composition databases and dietary intake studies. The materials support implementation of harmonised food analysis protocols globally.

Key measures

Mass fractions of vitamin D3 (0.498–49.5 μg/kg), vitamin D2, and 25(OH)D3 (0.53–12.1 μg/kg) in five food matrices (meat homogenate, whole milk powder, bovine liver, whole egg powder, soy milk) measured by isotope dilution LC–MS/MS

Outcomes reported

NIST established certified reference values for vitamin D2, vitamin D3, and 25-hydroxyvitamin D3 in five food-matrix Standard Reference Materials using isotope dilution LC–MS/MS. The study provides reference materials to support analytical method validation and inter-laboratory comparability of vitamin D measurements.
